5. 내 용 : This presentation will describe the development and application of novel microfluidic devices for neuroscience and stem cell research.. Microfluidic systems offer several advantages over traditional cell culture models due to its ability to control precise microenvironment in space and time when applied to cell biology problems. This presentation will describe (1) gradient-generating microfluidic devices that are used to understand axon guidance, cell migration and stem-cell differentiation/differentiation and (2) a microfluidic neuronal culture platform that combines microfabrication and surface micropatterning approaches to create an experimental platform for Alzheimer’s Disease, traumatic brain injury, and spinal cord injury research. Demonstration of axotomy, isolation of pure axonal fraction and biochemical analysis, and axonal transport with relevance to neurodegenerative diseases research will be highlighted. The ability to direct sites of neuronal attachment and orientation of axon outgrowth by micropatterning techniques, combined with fluidically isolated compartments within the culture area offer significant advantages over conventional methods for manipulating distinct cellular microenvironments.
