5. 내 용 : Progressive microglial accumulation at amyloid-β (Aβ) plaques is a well-established signature of the pathology of Alzheimer’s disease. To understand the distinct roles of Aβ on microglial accumulation, we provided controlled environment of week-long lasting gradients of soluble Aβ and patterns of surface-bound Aβ. We found that human microglia chemotaxis in gradients of soluble Aβ42 was unusually most effective at two distinct concentrations. We uncovered that chemotaxis at lower concentrations was enhanced by Aβ-induced microglial production of MCP-1. Microglial migration was inhibited by surface-bound Aβ42. We envision that the understanding on microglial migration can provide the insights for the treatment of AD.
To better understand the roles of BBB in various diseases and for screening CNS-targeting drugs, we have designed a platform that replicated critical mechanical and functional features of the BBB in 3D. The tightness of our BBB was validated by observing localized membrane proteins of VE-cadherin and ZO-1 along cellular boundary and blocking transmigration of innate immune cells through our BBB. The tight sealing of our BBB model could be disrupted under the treatment of TNF-α. We envision that our model can be easily integrated with other components to add more functionality for screening drug candidates.
6. 약 력 :
Dr. Hansang Cho earned his Ph. D in Bioengineering in 2010 from University of California, Berkeley. He was a team manager developing artificial hearts at Biomedlab. Co. during 1999 and 2003, a researcher at KIST during 2003 to 2010, at Lawrence Livermore National Laboratory during 2005 to 2010. He is currently a research fellow at Harvard Medical School/MGH since 2010. Dr. Cho is currently developing brain-on chips for neurodegeneration, and chemotaxtic platforms for inflammation.
